Provides technical leadership for in vitro ADME studies to support ongoing projects. Will design, coordinate, and execute in vitro ADME studies to support small molecule structure-based drug discovery and development for treatment of neurodegenerative diseases; establish, design, validate and conduct in vitro experiments using cell culture techniques and maintain immortalized, transfected cell lines; maintain, make improvements in database systems for use in calculating cellular permeability and the uptake of compounds; establish and validate assays for drug transporter interaction estimation in membrane vesicles; perform LC-MS/MS analysis of plasma, blood and brain samples from pharmacokinetic studies, including sample preparation and LC-MS/MS operation and method development; perform and analyze data from in vitro enzyme kinetic studies for pharmacokinetic and drug metabolism studies, estimate kinetic parameters IC50, Ki, Kinact; take lead on troubleshooting complex issues, utilize scientific literature to implement advancements in the current procedures; lead in-house permeability and transporter interaction studies for developmental stage compounds, write and execute study protocols, compile findings in the form of formal report; collaborate and coordinate with CRO for preclinical studies; organize and review experimental write-ups and study reports from CROs; utilize understanding of GLP principals; analyze, interpret data, incorporate results into scientific projects and communicate preclinical study data with project teams; utilize knowledge of drug discovery screening and high-throughput methods, as well drug metabolism principles and conduct hands-on research to acquire, implement, and execute assays to support our lead optimization activities; help guide structure activity relationship for the ongoing projects; train coworkers on the novel in vitro ADME assays; serve as authorized radiochemical user, supervise hands-on radiochemical user training, help maintain radiochemical use facility; exercise independent judgment in selecting methods, techniques and evaluation criteria for obtaining results.
Bachelor's degree in Biochemistry, Molecular Biology, or Biological Sciences (willing to accept foreign education equivalent) as well as 8 years of experience in Job Offered or 8 years of experience in designing and conducting in vitro permeability, transporter experiments, in vitro drug metabolism studies and LC/MS analysis. Or, alternatively, a Master's degree and 5 years of experience as noted above. Experience must include: Demonstrated Expertise (?DE?) culturing, maintaining Caco-2, MDCK cells and performing permeability studies using Caco-2, LLC-PK1 cells, including experience working with GLP practices for permeability and in vitro metabolism studies; DE performing in vitro transporter interaction studies in Caco-2 cells, LLC-PK1 cells, MDCK cells, membrane vesicles, oocytes, hepatocytes and kinetic parameters IC50, Km estimation in these models including use of radiolabeled compounds/radiochemical detection, membrane protein extraction and qualitative estimation of transporter proteins using western blotting; DE performing in vitro ADME studies including CYP/UGT/FMO reaction phenotyping, competitive inhibition, time dependent inhibition, kinetic parameters IC50, Ki/Kinact estimation, metabolic stability estimation in hepatocytes, microsomes, recombinant drug metabolizing enzymes and use of Tecan automation for these assays; DE in bioanalysis, including the preparation of blood, plasma and brain samples for LC-MS/MS analysis using solid phase extraction, liquid-liquid extraction and protein precipitation as well as LC-MS/MS operation, method development and data analysis using 3200 Q TRAP, API 4000 and QTRAP 5500, and working with software packages to include AB Sciex Analyst, Waters Empower, XLfit, SigmaPlot, E-Workbook, Dotmatics and Vortex.
